Isolation of Novel Sesquiterpene Lactone from Vernonia Colorata

Species: Vernonia colorata Region: Southwestern Nigeria August 2025

Significance

First isolation of a novel sesquiterpene lactone with demonstrated anti-inflammatory activity from V. colorata leaves, supporting traditional use for treating inflammatory conditions.

Discovery Details

During routine phytochemical screening of Vernonia colorata leaf extracts collected from Ogun State, our team isolated a previously undescribed sesquiterpene lactone compound, designated IRL-VC-01. The compound was characterized using 1D and 2D NMR spectroscopy, high-resolution mass spectrometry (HR-MS), and X-ray crystallography.

Bioactivity

Preliminary bioassays demonstrated significant anti-inflammatory activity in the carrageenan-induced paw edema model, with IRL-VC-01 showing dose-dependent inhibition of inflammatory markers (COX-2, TNF-α, IL-6) at concentrations comparable to indomethacin.

Traditional Context

V. colorata has been used by Yoruba traditional healers in southwestern Nigeria for centuries to treat a range of inflammatory conditions, including joint pain and skin irritation. This discovery provides molecular evidence supporting these traditional applications.

Next Steps

We are currently conducting structure-activity relationship (SAR) studies and scaling up extraction to support further pharmacological evaluation. A full characterization paper is in preparation for submission to the Journal of Natural Products.
